The experiment could lay the foundation for fundamentally new methods of treating acute liver failure, which annually kills more than 40 thousand people in the United States alone.

An experiment on the cultivation and implantation of an artificial analogue of the human liver was carried out by groups of scientists from the USA, Brazil, Japan and China, the results of their work were published by the researchers in the journal Cell Reports. The researchers emphasize that a lot of work remains to be done before this method can be used for clinical purposes, but if this happens, world medicine will receive a full-fledged alternative to donor liver transplantation. Today this procedure is very expensive, and moreover, most patients die without waiting for a donor organ.

“The long-term goal of our work is to learn how to create artificial organs that can replace donor organs. Our work can help with transplantation in the near future. For example, in case of acute liver failure, it will be possible to take the load off the liver rather than replace it with a whole donor organ,”says Alejandro Soto-Gutiérrez, one of the authors of the experiment, a pathologist at the University of Pittsburgh.

To grow a mini-liver, the researchers took skin cells from volunteers and reprogrammed them to return them to a state of stem cells. With the help of hormones and other chemicals, they were turned into liver cells, which were cultured in the laboratory. As a result, scientists obtained a full-fledged liver tissue with blood vessels and bile ducts. And although it takes at least two years for a human liver to "mature" from the moment of birth, the researchers were able to grow a miniature analogue of the organ in a few weeks, after which it was implanted into five rats.

The experiment was successful: the liver took root and began to produce bile and cleanse the blood of toxins. This is evidenced by the results of the autopsy after killing the animals four days after implantation.

Although the artificial liver functioned well enough to keep the rats alive, studies showed insufficient blood flow to the organ. This suggests that there are difficulties in connecting an artificial organ with the vascular network of an animal, scientists emphasize.

However, the work carried out will allow in the near future to use mini-organs to study various diseases and test treatment options.

“I think this is a very important step, because now we know that it is possible to create a whole functioning organ from one cell,” said Soto Gutierrez.
